Same receptors, different interactions states.

One of the major challenges in GPCR pharmacology is that receptors do not exist in a single static state.Coupling to intracellular proteins, receptor trafficking, internalization, and recycling can all alter the observed interaction between a ligand and its target.

In our recent publication, we used LigandTracer and InteractionMap to study the interaction between 177Lu-DOTATATE and the GPCR SSTR2 in real time on living cells. The kinetic data revealed multiple interaction populations with distinct binding stabilities, enabling us to connect observed affinity differences directly to receptor biology.

Key findings include

  • Strong-affinity interactions associated with G protein-coupled receptors
  • Weak-affinity interactions associated with uncoupled receptors
  • Contributions from receptor internalization and recycling
  • Time-dependent shifts in receptor-state populations following prolonged stimulation

The study demonstrates how real-time cell-based assays can reveal layers of biology that remain hidden in conventional endpoint assays.

For GPCR research, understanding why interactions occur can be just as important as measuring how strong they bind.
