Cell-cell interaction dynamics and half-life quantification using COE-based fluorescent probes and the LigandTracer platform.
Results from a proof-of-concept study between Adamastor Bioservices and Acoerela were recently presented at ISCT Singapore, demonstrating the use of COE-based fluorescent probes and
LigandTracer to study the binding dynamics of cell-cell interactions. Real-time information on how immune cells engage, form a complex, and eventually disengage is key to understand downstream biological outcomes.
The LigandTracer platform is opening new possibilities for studying cell-cell interactions through:
- A membrane-associated fluorescent labelling workflow for living cells using COE-based Aco Dyes™.
- Quantification of half-life and affinity in macrophages-tumor cell interactions
Tumor cells were immobilized on a Petri dish and opsonized with IgG antibodies. LigandTracer was used to study the interaction with COE-TAB labeled macrophages. The results show that the macrophages interacted with a 7.3e5 cells/ml affinity (KD), i.e., the concentration required to achieve 50% target cell binding. The interaction showed an association rate (ka) of of 2.2e-6 ml·cell-1·h-1 and a dissociation rate (kd) of 4.5e-4 s-1, corresponding to a half-life of half an hour.
COEs are water-soluble, fluorogenic, lipid-bilayer-spanning probes that remain strongly associated with lipid membranes. Their membrane association provides bright fluorescent signals suitable for tracking labelled cells over time, while their water solubility supports straightforward preparation in aqueous media.
